چکیده

Sites of renal Mg transport were identified in seawater killifish ( Fundulus heteroclitus) using a Cameca model IMS-3f ion microscope. Killifish were given an intraperitoneal injection of the stable isotope26Mg (99.5% enrichment) to stimulate and trace renal Mg excretion. We identified two sites of26Mg transport in frozen freeze-dried cryosections of kidney: the proximal tubule, known to secrete Mg, and the collecting duct, heretofore not known to handle Mg. In epithelial cells of the proximal tubule, the punctate distribution of injected 26Mg suggests transcytotic excretion of Mg in bound form. In collecting ducts, a subpopulation of Mg/Ca-rich cells was identified with high accumulations of injected 26Mg. Here, the punctate distribution of26Mg decreased from the apical to the basal region of the cells, revealing a transcytotic gradient of apparently bound Mg. Since proximal tubules of fish are implicated with Mg secretion, Mg/Ca-rich cells in the collecting duct may reabsorb Mg, thereby providing the usual two-step of renal regulation, now also for Mg.

Hydrochemistry and stable isotopes study of the precipitation at Haraz Basin, north of Iran

Stable isotopes (Deuterium and Oxygen-18) have a broad application in many of water related sciences. Precipitation is the most important input to the water cycle which shows considrable spatial and temporal variability in its isotope content. To study the precipitation isotope content, 51 samples from August 2015 to July 2016 have been collected mainly on a monthly basis with few events based ...

Determining an accurate method to estimate GFR in renal transplant recipients with stable serum creatinine levels

Introduction:Detecting renal allograft dysfunction early will allow timely diagnosis and treatment. There is no objective recommendation by national kidney societies for glomerular filtration rate (eGFR) estimation in post-transplant setting. 99mTc-DTPA Technetium-99m Diethylene triamine penta acetic acid) renogram can identify early renal dysfunction much before ser...

Sub-cellular localization of a N-labelled peptide vector using NanoSIMS imaging

Dynamic SIMS imaging is proposed to map sub-cellular distributions of isotopically labelled, exogenous compounds. NanoSIMS imaging allows the characterisation of the intracellular transport pathways of exogenous molecules, including peptide vectors employed in innovative therapies, using stable isotopes as molecular markers to detect the compound of interest. Shiga toxin B-subunit (STxB) was ch...

Towards a Guided Regeneration of Renal Tubules at a Polyester Interstitium

Stem/progenitor cells are promising candidates for a therapy of renal failure. However, sound knowledge about implantation and regeneration is lacking. Therefore, mechanisms leading from stem/progenitor cells into tubules are under research. Renal stem/progenitor cells were isolated from neonatal rabbit kidney and mounted between layers of polyester fleece. It creates an artificial interstitium...

Cloning of rainbow trout SLC26A1: involvement in renal sulfate secretion.

The kidney plays an important role in ion regulation in both freshwater and seawater fish. However, ion transport mechanisms in the teleost kidney are poorly understood, especially at the molecular level. We have cloned a kidney-specific SLC26 sulfate/anion exchanger from rainbow trout (Oncorhynchus mykiss) that is homologous to the mammalian SLC26A1 (Sat-1). Excretion of excess plasma sulfate ...
